At South Pointe High School in Rock Hill, South Carolina, Gilmore participated in football, basketball, and track. He was a quarterback and defensive back in football, and as a senior, he led his squad to the SCHSL AAAA Division II championship and a perfect 15-0 record.
He played college football for South Carolina, and the Buffalo Bills selected him as the tenth overall choice in the 2012 NFL Draft.
Before joining the New England Patriots, Gilmore spent his first five seasons playing with the Buffalo Bills, making the Pro Bowl in 2016.
